Description:

The Ebola virus is an enveloped, non-segmented, negative-strand RNA virus, belonging to the filoviridae family. Totally, five species of the virus have been identified, including Zaire ebolavirus, Sudan ebolavirus(Sudan virus), Bundibugyo ebolavirus (Bundibugyo virus), Taϊ Forest ebolavirus (Taϊ Forest virus), and Reston ebolavirus (Reston virus). The full-length transmembrane glycoprotein (GP1,2), one of the structural proteins encoded by Ebola viruses, is the virus surface protein and forms trimeric spikes that facilitate virus entry by mediating receptor binding and membrane fusion. GP1 and GP2 are the disulfide-linked subunits formed during the processing in the endoplasmic reticulum and Golgi apparatus , which are furin-cleavage products of the full-length GP polyprotein precursor. GP trimerization is mediated through multiple GP1-GP2 and GP2-GP2 contacts. GP1 is responsible for cell attachment, while GP2 is responsible for fusion of viral and host cell membranes. GP1 is composed of a single domain ( ~65 Å×30 Å ×30 Å) arranged in the topology, and can be further subdivided into the (I) base, (II) head and (III) glycan cap regions. GP2 contains the internal fusion loop and the heptad repeat regions, HR1 and HR2.
