Description:

The glycoprotein (gp) 120, an essential proportion of the envelope spikes that decorate the surface of HIV-1 as well as a major target for neutralizing antibodies, is anchored to the viral surface by the transmembrane unit, gp41. The trimeric envelope glycoproteins (Env), also called gp160, are heterodimers of gp41 and gp120. Initially, gp120 and gp41 are synthesized as a single gp160 polypeptide that is subsequently cleaved to generate the noncovalently associated gp120/gp41 complex. Gp120 is composed of five variable regions, or loops interspersed with five conserved regions. Gp120 is a globular structure that folds to form an inner domain, an outer domain, and a bridging sheet that links the inner and outer domains. The CD4 binding site is located in the pocket between the inner and outer domain and above the bridging sheet. All three domains are crucial for CD4 and coreceptor binding. Glycosylation exists over much of the surface of the protein, particularly on the outer domain and the immunosilent face that contains the V4 and V5 variable loops. Gp140, a heterodimer with mass of approximately 140 kDa, is the ectodomain of Env. The glycoprotein gp140 is comprised of the whole gp120 component and approximately 20 kDa of gp41, which are displayed on the surface of the viral membrane. Soluble versions of trimeric gp140, either cleaved or uncleaved, are being developed as immunogens to elicit a protective humoral immune response against HIV-1 infection.
